Unscheduled DMA Synthesis Induced by Procarcinogens in Suspensions and Primary Cultures of Hepatocytes on Collagen Membranes1

چکیده
Unscheduled DMA synthesis was induced by procarcinogens in freshly isolated suspensions and primary cul tures (6 days old) of hepatocytes on collagen membranes. Incorporation of ['HJthymidine in the presence of hydroxyurea was used to measure unscheduled DMA synthesis. When hepatocellular DNA was isolated on cesium chlo ride gradients, significant levels of unscheduled DNA synthesis were measured. Similar concentrations of procarcinogens elicited higher levels of unscheduled DNA synthesis in hepatocellular suspensions than in primary cultures. The results demonstrate that hepatocytes cul tured on collagen membranes can metabolize chemical carcinogens. Suspensions of freshly isolated hepato cytes, however, are more active in procarcinogen metab olism than are those of primary cultures. The selective advantages of the two systems of hepatocytes can be utilized for the establishment of short-term in vitro screen ing systems of mutagens and carcinogens.

The procarcinogen, 2-acetylaminofluorene, the direct-acting carcinogen, methyl methanesulfonate, and two other hepatocarcinogens, thioacetamide and urethan, were tested for their ability to elicit unscheduled DNA synthesis in adult rat hepatocytes maintained in primary culture on collagen gel-nylon mesh. The carcinogens, dissolved in dimethyl sulfoxide were added to 6-hr or to 28-hr cultures al...

Unscheduled DNA synthesis was observed in primary rat liver cell cultures treated with members of five different classes of chemical procarcinogens requiring enzymatic activation as well as with a direct-acting carcinogen. In total, ten carcinogens and one related analog not commonly accepted as carcinogenic were active, while one weak carcinogen and four noncarcinogens were inactive. The produ...

Heretofore, unscheduled DMA synthesis (UDS) induced by DMA-damaging chemicals has been measured in nonreplicating human fibroblasts by autoradiographic meth ods that are not readily applicable to organotypic epithe lial cell cultures. In order to evaluate the range of chemi cal sensitivity and DNA repair response of human skin epithelial cells, we have developed a novel semiquantitative in vitr...

A method has been developed for the rapid preparation of single-cell suspensions from rat hepatocyte primary cultures on collagen substratum. Hepatocytes were adequately dissociated into single cells when the cultures were first treated with a combination of trypsin and ethylenediaminetetraacetic acid (EDTA) and then with collagenase. However, when the order was reversed, hepatocytes were inade...

The effect of certain procarcinogens, among which demethylnitrosamine (DMN) is included, has been difficult to detect in several short-term assays. An alternative system, in which DMN effects could be easily quantitated, might be useful in studies of chemical carcinogenesis and environmental contamination. To develop such a system, we tested the possibility of measuring the amount of breakage p...
